No, it wasn't. You will find no evidence in existence that the Germans classified tanks by gun caliber. I don't know how in the world that long-debunked theory from the eighties got started.
The Pz.Kpfw III was a medium tank, and it used a 37-mm gun at one point. Then a 50-mm gun. Then the Pz.Kpfw IV and Pz.Kpve V come along with 75-mm guns. In fact, the Pz.Kpfw IV with a 75-mm gun was originally intended to be a 'heavy' tank.
Obviously, gun size wasn't by any means a factor in their classification.
