I think it's because PonE usually features in stories set in basically modern-day Earth (one spaghetti-flavoured pill richer). Being a pone transported to Equestria is great, being a pone as part of some widespread trans-species program is great, but becoming the permanent pone through illegal drug abuse would be kind of detrimental to life in society.
Now while I'm sure some people around here would happily take two pills and go live innawoods as an outcast, many others would be more hesitant (I know for a fact double-dosing would be absolutely inevitable for me eventually, but I'd still be worried enough about my current life to take it slow at first). Also as far as stories go, it's more interesting to see the protag progressively lose themselves in it, than "This is bob, he really wants to be a pony. He takes two pills with no regards to the consequences. He is now overjoyed, not caring about anything else. The end."

Can't think of any off the top of my head, if someone knows of any, would be interesting to read. There's maybe Keyboard Masher who took two by accident, but I don't know of any where the anon takes them intentionally.

Maybe a good plot would be to have happy-go-lucky average joe anon just dive headfirst into "yay pony! yay hooves and tail and ears!" but then have to deal with the reality of surviving as a slightly-less-than-human social outcast until he finds a transformee commune, and in the process learns to take life more seriously and commit to responsibilities leading to starting a happy pony family

Honestly, however you want. The canonical answer is "anon is a blank slate self-insert" but in 99% of stories he's not a blank slate, he's got at least a coherent characterisation. He's often got only a vague background, but I think that's mostly because he's often used for relatively quick, low-setup stories, as a "pre-made" character you can just grab and go (like if you wanna write an HiE but don't wanna concern yourself with how the human actually arrived in equestria - you can just write an AiE instead and nobody will bat an eye).
But there's also plenty of stories that give more of a background or backstory, or provide well-developed characterisations. So to answer your question, develop your anon's character in a way to best serve your story. You can have a dead end loser pizza delivery anon who's a suttering sperg, just as much as you can have a nuclear technician anon, or a hobo anon who's lost one eye and likes to write political satire poetry in public stalls. If it fits your story, you're good to go.
